Monoclonal antibodies are among the most significant biological tools used in medicine and biology that have revolutionized the field of diagnostics, therapeutics, and targeted drug delivery systems for many diseases[1]
Among them, rabbit monoclonal antibodies have attracted significant attention for having high affinity and specificity[2]. We describe a robust platform to generate anti-IDs using rabbit single B cell sorting-culture and cloning technology by immunizing rabbits with therapeutic drug Fab fragment and sorting complementarity determining regions specific B cells using designed framework control as a negative gate to exclude non-CDRs-specific B cells. The supernatants of cultured B cells were subsequently screened for binding to drug-molecule by enzyme-linked immunosorbent assay and the positive hits of B cell lysates were selected for cloning of their immunoglobulin G (IgG) variable regions[3].

Antigen Preparation
Customers may provide highly purified (>90%) protein or peptide antigens that are ready for injection. Alternatively, we provide services for protein production, peptide design, peptide synthesis, and peptide conjugation to carrier protein for use as the immunogen.
Immune Animal
1.Each rabbit was immunized by subcutaneous (SC) and intradermal (ID) injections at multiple sites according to reagent ratio. After the initial immunization, the immunization was enhanced three times by SC injection.
2.After surgery, the rabbits recovered from anesthesia and were continuously monitored for signs of lethargy until they were fully mobile and active. Milliliters of blood were collected from the middle auricular artery with a butterfly needle.
Cell Screening
Rabbit peripheral blood mononuclear cells were isolated by whole blood density centrifugation of lymphocytes. IgG B cells were screened negative.
Antibody titers were measured
The antipolyclonal serum titer was determined by standard ELISA.
