    Phosphopeptides are produced by the specific incorporation of protected phospho-amino acids or the "post-assembly" peptide phosphorylation at serine, threonine or tyrosine residues. In cells, about one-third of proteins are thought to be modified by phosphorylation.Phosphorylation of serine (Ser), threonine (Thr) and tyrosine (Tyr) is a reversible protein modification process that is involved in the regulation of numerous cellular activities, such as receptor signaling, protein association and segmentation, activation or inhibition of protein function, and even cell survival.

    Biotyscience can provide customers with peptide synthesis and peptide modification services. Phosphopeptides play an important role in life processes and are phosphorylated at the position of peptides such as Ser, Thr, and Tyr. We offer both monophosphorylated and polyphosphorylated peptide services, and we are currently able to offer peptides modified at four phosphorylated sites.

    Details

    The post-phosphorylation method is used to selectively remove the side chain protection group of the amino acid to be labeled after the synthesis of the peptide. For Tyr, Thr can directly react with amino acids that are not protected by the side chain, while Ser can be quantitatively removed using Fmoc-Ser (trt) under 1% TFA/DCM. After phosphorylation, bibenzylphosphamide and tetrazolium are used to form phosphamide and tetrazolium active intermediates, which are connected to hydroxyl group, and then oxidized under peroxic acid to form phosphoyl group to complete the reaction.The final product is provided with corresponding mass spectrometry, HPLC chromatogram for purity analysis, and COA.

    4.3 Phosphorylation of 2 sites, 3 sites, 4 sites, and 5 sites can be performed simultaneously.
