The post-phosphorylation method is used to selectively remove the side chain protection group of the amino acid to be labeled after the synthesis of the peptide. For Tyr, Thr can directly react with amino acids that are not protected by the side chain, while Ser can be quantitatively removed using Fmoc-Ser (trt) under 1% TFA/DCM. After phosphorylation, bibenzylphosphamide and tetrazolium are used to form phosphamide and tetrazolium active intermediates, which are connected to hydroxyl group, and then oxidized under peroxic acid to form phosphoyl group to complete the reaction.The final product is provided with corresponding mass spectrometry, HPLC chromatogram for purity analysis, and COA.
